Kuhle Wampe takes place in early-1930s Berlin. The film begins with a montage of newspaper headlines describing steadily-rising unemployment figures. This is followed by scenes of a young man looking for work in the city and the family discussing the unpaid back rent. The young man, brother of the protagonist Anni, removes his wristwatch and throws himself from a window out of despair. Shortly thereafter his family is evicted from their apartment. Now homeless, the family moves into a garden colony of sorts with the name “Kuhle Wampe.”

The central character of the play, Fedor Protasov, is tormented by the belief that his wife Liza has never really chosen between him and the more conventional Victor Karenin, a rival for her hand. He wants to kill himself, but doesn't have the nerve. Running away from his life, he first falls in with Gypsies, and into a sexual relationship with a Gypsy singer, Masha.

Mr. Blackwell discovers a relic that informs him about Blythe (as Ayesha, or "She-Who-Must-Be-Obeyed"), who loved his father and others in the ancestral line. Blackwell accompanies pal Heinrich George and handyman Tom Reynolds to Arabia.

Paul and Peter share a garret flat. Paul is smitten with Anni, but her father forbids their match. Then Paul inherits a fortune—on one condition: he mustn’t marry “a girl.” Desperate, they scheme a sham marriage: Peter weds Anni so she’s no longer “a girl,” allowing Paul to claim his inheritance. Jealous at first, Paul watches as Peter and Anni wed, until, after their quick divorce, Paul finally wins Anni’s hand for real.

Mutter Krause, her daughter Erna and her son Paul live in a tenement in the poorer section of Berlin's Wedding district. Along with them lives "the Tenant", his soon-to-be bride Friede, who works as a prostitute, and her child whose name isn't revealed in the film. Mutter Krause is a quiet, long-suffering old woman who earns what little she can delivering newspapers. However, Paul is an alcoholic and spends all her money on drink. Mutter Krause can't pay back the money she owes the man whose newspapers she delivered and he accuses her of stealing and threatens her with arrest. Mutter Krause must then pawn her last valuable possession, a treasured memento of her late husband. Paul then breaks into the same pawn shop. He gets away but is later arrested. Meanwhile, Erna begins dating a young man with Communist views, who turns Erna to Communism and also helps her earn the money her mother needs by more honest means.

A sordid tale of poverty and greed set within a quayside milieu of crime and prostitution, the narrative centres on the quest for a sparkling pearl necklace stolen by a beggar under the gaze of a prostitute, who persuades her unemployed friend to steal it back, with tragic consequences. The story unfolds in flashback, without irony or a hint of redemption: life simply goes on.

The film is based on real events and reveals the tragic episodes from the life of the Austrian biologist scientist-materialist Paul Kammerer (1880-1926), hunted by regressive scientists and Catholic reactionaries who committed suicide.
